Inhibitory effect of panicoin on platelet activation and its mechanism |

Abstract: |
Objective: To explore the inhibitory pathway and the possible mechanism of panicoin on platelet activation.Methods: The effect of panicoin on platelet aggregation, thromboxane B2 release, P-selectin (GMP-140) count, levels of cAMP and cGMP induced by ADP, epinephrine, arachidonic acid or thrombin were observed.Results: Panicoin could inhibit significantly platelet aggregation induced by ADP, arachidonic acid and epinephrine in a dose-dependent manner, thrombin induced GMP-140 on platelet surface and TXB2 release, and increase the platelet cAMP level, but showed no effect on cGMP level.Conclusion: Panicoin shows a potent inhibition on platelet activation and antithrombotic effect. The mechanisms seems to be related with elevation of platelet cAMP. |
