      本文报告赤芍成分和其衍生物801、802对 ADP 和胶原诱导的血小板聚集性均有不同程度的抑制作用,同时,还降低线粒体氧耗量(S3)、磷/氧(ADP/O)和呼吸控制比(RCI)。并发现上述药物的构效关系和量效关系,证实了血小板聚集性与能量代谢的相互关系。
英文摘要:
      Radix Paeoniae Rubra,a common traditional Chinese drug with the action of promoting blood circulation and removing blood stasis,has been shown to be able to inhibit platelet aggregation,act vate fibrinolysin,promote thrombolysis.This paper describes the influence of the extract of Radix Paeoniae Rubra and its two derivatives (801,802)on platelet aggregation induced by ADP or collegen and rat liver mitochondria oxidative phosphorylation in an attempt to explore the relationship between the anti-platelet aggregation effect and energy metabolism. The experimental results show that:(1)The extract of Radix Paeoniae Rubra and 801,802 can inhibit platelet aggregation induced by ADP or coltegen in different degrees,counteract platelet aggregation induced by ADP and delay occurrence of platelet aggregation induced by collegen.The order of effectiveness of anti-platelet aggregation is:802>801>extract of Radix Paeoniae Rubra(final concentration 5×10-4-5×10-3M).The effect of these compounds show that there are dose-response and structure-response behaviours.(2)All these compounds can decrease rat liver mitochondria oxygen consumption(S3),respiratory control index(S3/S4)and ADP/0 ratio in different degrees to regulate energy prod uction.The order of the effectiveness of these drugs in this respect is the same as the potency of their anti-platelet aggregation action.Therefore,it could be assumed that the regulation ofenergy metabolism by drugs is one of the ways to improve the physiological function of platelets.(3)The biochemical pharmacological action of the extract of Radix Paeoniae Rubra and its derivatives are related with the number of carbon increase,the hydrophobic properties will also increase,the effects of inhibition are more significant.These preliminary results help to throw more light on the mechanism of anti-aggregation of t he Chinese medicinal herbs.
