      目的 :观察小鼠烧伤前后肠道免疫分泌的变化和大黄汤治疗对正常小鼠和烧伤小鼠肠道免疫分泌的促进作用 ;探讨大黄保护肠粘膜屏障的作用机理。方法 :实验用BALB/c小鼠。分为 4组 :1组为对照组 (n =8) ;2组为正常小鼠饲喂 10 %大黄液 12ml/kg ,每 6h 1次 (n =11) ;3组为 30 %的全厚背部皮肤烧伤 ,不治疗 (n =7) ;4组为全厚皮肤烧伤 ,同 3组 ,大黄治疗同 2组 (n =9)。 2 4h后处死动物 ,灌洗肠道 ,收集肠液透析离心后作IgA检测和总蛋白 ,补体C3,高密度脂蛋白 (HDL)测定。结果 :1、2、3和 4组小鼠肠液中的IgA(mg/L)分别为 93 5± 13 7、15 0 8± 4 4 6、4 3 4± 2 1 5及 5 9 8± 19 3,说明在烧伤后肠液中IgA明显减少 (P <0 0 5 )。而应用大黄汤治疗后 ,无论正常小鼠和烧伤小鼠的肠道IgA均增加 (P <0 0 5 ) ,其他免疫相关物质如总蛋白、补体C3和HDL大黄治疗后也有显著增加。结论 :烧伤后小鼠肠液中的IgA明显减少。而大黄无论对正常小鼠和烧伤小鼠均具有明显的促进肠道免疫分泌的效果。这一作用是大黄的粘膜屏障保护作用的重要机制之一。
英文摘要:
      To explore the therapeutic mechanism of rhubarb (sennosides) in protecting intestinal muco membranous barrier by observing the change of intestinal immune associated secretion (IIAS) in mice before and after burn and the enhancing effect of rhubarb on it in healthy and in burn mice. Methods: Bal b/c mice were divided into 4 groups. Group A (n=8), untreated healthy mice; Group B (n=11), healthy mice treated with 10% rhubarb decoction 12 ml/kg, once every 6 hrs; Group C (n=7), the mice with 30% back full thickness burn and no treatment was given; Group D (n=9), the mice with 30% back full thickness burn and treated with 10% rhubarb decoction 12 ml/kg, once every 6 hrs. The animals were killed 24 hrs after burn, the intestinal juice was collected after intestinal lavage, and centrifuged for determination of IgA, total protein, C 3 and high density lipoprotein. Results: IgA content in intestinal juice of Group A, B, C and D were 93 5±13 7mg/L, 150 8±44 6mg/L, 43 4±21 5mg/L and 59 8±19 3mg/L respectively, suggesting that it was significantly reduced by burn (P<0 05), while rhubarb decoction could increase it in either healthy or burn mice (P<0 05). The other immune associated substance, such as total protein, C 3 and high density lipoprotein in intestinal juice were also increased significantly after rhubarb decoction treatment. Conclusion: Rhubarb could increase the reduced intestinal juice IgA content in mice caused by burn, it may be an important mechanism of rhubarb in protecting muco membranous barrier.
