      目的:探讨中药复方对MRL/lpr狼疮小鼠肾脏基因表达及Th1/Th2细胞因子比例的调节作用。方法:将狼疮小鼠随机分为两组。治疗组每天经饲料喂服中药复方,对照组进食正常鼠饲料,两组均自由饮水。观察小鼠存活率、蛋白尿阳性率及肾脏病理改变,将小鼠肾组织mRNA与点有8192条小鼠基因的芯片进行杂交,RT-PCR法检测肾组织干扰素γ(IFN-γ)和白细胞介素4(IL-4)的表达。结果:存活率治疗组为81.3%,对照组为51.2%;蛋白尿阳性率治疗组为40%,对照组为80%;治疗组肾小球与肾小血管病变较轻。芯片结果显示两组间差异表达(2倍以上)的基因共279条。RT-PCR结果显示治疗组肾组织中Th1/Th2细胞因子(IFN-γ/IL-4)比例增加(与对照组比较)。结论:中药复方能改善狼疮小鼠肾脏病变,调节肾组织多个系统的基因表达,纠正Th1/Th2细胞因子的比例失衡。

      Objective: To explore the effect of Chinese compound recipe (CCR) on gene expression and its regulatory effect on Thl/Th2 cell ratio in kidney of autoimmune MRL/lpr mice. Methods: Autoimmune MRL/lpr mice, aged 8 weeks were divided into two groups. The treated group was fed by the forage added 60 g/kg CCR, while the control group fed by normal forage, all taking water freely. The survival rate, positive proteinuria rate, change of renal pathology were observed, the mRNA of mice’s renal tissue was hybridized with the gene chip having 8192 mice genes, and the expressions of interferon-γ (IFN-γ) and interleukin-4 (IL-4) were determined adopting RT-PCR method. Results: In the treated group the survival rate was 81.3% and the positive proteinuria rate 40% , while in the control group, it was 51. 2% and 80% respectively. The pathological changes of renal glomeruli and small vessels were milder in the treated group than those in the control group. Gene chip hybridization showed that between the two groups, 279 genes were differently expressed for over 2 times, and result of RT-PCR showed that the Thl/Th2 cytokine ratio (IFN-γ/IL-4) was higher in the treated group than that in the control group. Conclusion: CCR could improve the renal pathological changes, regulate the gene expression of multiple systems of renal tissue, correct the imbalance of Thl/Th2 cytokine in kidney of MRL/lpr mice.
