      目的 :观察骨松宝对体外培养成骨细胞增殖和代谢的影响。方法 :给 18月龄老年大鼠灌胃骨松宝 1 5 g/kg体重 ,每日 2次 ,连续 3天 ,末次灌胃后 1h取血分离血清 ,用D890 0培养基分别配成含药鼠血清7 5 %和 15 %的培养基 ,并用于培养成骨细胞 2 4h ,同时以加有 7 5 %和 15 %未用药老龄大鼠血清的D890 0培养基、含 2 0 μmol/L氟化钠 (NaF)的D890 0无血清培养基及D890 0无血清培养基作对照 ,用MTT法检测细胞增殖 ,并测定培养基上清液中Ca2 + 浓度及碱性磷酸酶 (ALP)的含量变化。结果 :与加有相同浓度鼠血清的D890 0培养基组、D890 0培养基加NaF组和D890 0培养基组比较 ,含药鼠血清组的成骨细胞增殖明显 ,差异有显著性 (P <0 0 1) ,培养基中的Ca2 + 消耗量明显增加 (P <0 0 5 ,P <0 0 1) ,ALP浓度明显升高 (P <0 0 5 ,P <0 0 1)。结论 :骨松宝能促进成骨细胞DNA合成和提高对Ca2 + 的利用 ,促进成骨细胞的生长增殖。

      Objective:To observe the effect of Gusongbao (GSB) on proliferation and metabolism of osteoblast cultured in vitro. Methods:Old rats, aged 18 months, were given GSB 1.5 g/kg, twice a day for 3 days by intragastric perfusion. Blood of the rats was collected 1 hr after the final perfusion to isolate serum for preparing, with D8900 medium, the culture media containing 7.5% or 15% GSB, which was used to culture osteoblast for 24 hrs. Besides, D8900 media containing 7.5% or 15% old rats′serum without medication, containing 20 μmol/L sodium fluoride, and simple D8900 medium were prepared for control. The cell proliferation was detected by MTT method, and the changes of Ca 2+ concentration and ALP content in supernatant of culture were also observed. Results:The osteoblast proliferation cultured in GSB serum containing medium was significantly increased than those cultured in the other control media (P<0 01), at the same time, the Ca 2+ consumption increased and the ALP content elevated significantly (P<0 05, P<0 01). Conclusion:GSB could promote the DNA synthesis, increase the utilization of Ca 2+ and accelerate the growth and proliferation of osteoblast.
