      目的研究丹参水溶性成分———丹参酚酸B镁对人主动脉内皮细胞生长增殖的影响和对自由基损伤内皮细胞的影响 ,以进一步了解丹参酚酸B镁的心血管药理作用机理。方法实验中使用第 3~ 6代的人主动脉内皮细胞 (humanaorticendothelialcell,HAEC) ,采用形态学和MTT法观察不同浓度药物对细胞增殖的影响 ;采用葡萄糖 -葡萄糖氧化酶造成的氧自由基损伤细胞模型观察药物的作用 ;透射电镜观察细胞超微结构的变化。结果丹参酚酸B镁 1 0mg/ml和 0 5mg/ml两个剂量对HACE有明显的细胞毒作用(P <0 0 1)。低于 0 2mg/ml的浓度对细胞增殖无不良影响 ;丹参酚酸B镁对自由基损伤细胞有明显的保护作用。电镜结果显示其可以保护线粒体等细胞超微结构。结论丹参酚酸B镁对HAEC细胞毒性较低 ;对自由基损伤细胞有明显的保护作用

      ObjectiveTo study the cardiovascular effects of magnesium lithospermate B (MLB), a water soluble extract from red sage root, on human aortic endothelial cells (HAECs) proliferation and in treating free radical injured endothelial cells, so as to further understand the cardiovascular pharmacological mechanism of MLB.MethodsHAECs of 3 6 passages were used in the experiment. MLB of different concentrations was used to treat the cells, and cell proliferation was observed using morphological and MTT method. The free radical injured model cell was made by glucose glucose oxidase method for observing the effect of MLB on its microstructure by transmission electron microscopy. ResultsMLB in concentration of 1.0 mg/ml and 0 5 mg/ml had significant cytotoxicity (P<0 01), when its concentration was lower than 0 2 mg/ml, the drug showed no adverse reaction on cell proliferation. MLB showed significantly cell protective effect against free radical injury, the effect in protecting ultrastructure of cells, such as mitochondria, could be demonstrated in transmission electron microscopy. ConclusionMLB showed a low cytotoxicity on HAECs, it could obviously protect cells from free radical injury.
