      目的探讨清开灵注射液对大肠杆菌脑膜炎脑损伤的保护作用机制。方法在采用氨苄青霉素治疗兔大肠杆菌脑膜炎模型的基础上,加用清开灵注射液辅助治疗。在注菌前、注菌后16、26h分别测定脑脊液中白细胞数、蛋白质含量和脑组织水、钠、钾、钙含量,并观察脑组织中基质金属蛋白酶9(MMP9)的表达。结果清开灵注射液辅助治疗后可明显减轻单纯抗生素治疗后细菌性脑膜炎模型兔脑脊液中白细胞数、蛋白质及脑组织水、钠、钙含量的增加和钾含量的下降,减弱脑组织MMP9的表达。结论清开灵注射液辅助治疗可防止单纯抗生素治疗细菌性脑膜炎时所引起的一过性炎症反应和脑损伤加重,其机制与钙拮抗作用和减弱脑组织中MMP9的表达有关。
英文摘要:
      ObjectiveTo explore the neuro-protective effect and mechanism of Qingkailing injection (QKL) against cerebral injury caused by E. coli-meningitis (CM).Methods The CM model rabbits were treated by ampicillin with QKL as adjuvant. The leukocyte count and protein content in cerebral spinal fluid (CSF), the contents of water, sodium, potassium and calcium in cerebral tissues were measured before, 16 h and 26 h after Bacillus coli injection respectively.The expression of matrix metalloproteinase-9 (MMP-9) was determined at the same time. ResultsAdjunctive treatment with QKL can not only inhibit the increase of leukocyte cells, protein content in CSF, and water, sodium, calcium content in cerebral tissues, but also the decrease of potassium content revealed during simple antibiotic treatment. It also can decrease the expression of MMP-9 in cerebral tissues of rabbits with CM. ConclusionAs an adjunctive treatment, QKL can prevent transient inflammatory reaction and aggravation of brain injury in CM induced by simple antibiotic treatment, its mechanisms might relate with calcium antagonism and attenuation of MMP-9 expression in brain tissues.
