      目的研究胡黄连苷Ⅱ对缺血再灌注后脑组织Toll样受体4(toll-like receptor4,TLR4)和核转录因子κB(nuclear transcription factorκB,NFκB)表达的影响。方法成年健康雌性Wistar大鼠60只,随机选取10只作为假手术组,其余应用线栓法建立大鼠大脑中动脉闭塞再灌注(MCAO/R)模型,将造模成功的30只大鼠随机分为阴性对照组、阳性对照组和胡黄连苷Ⅱ治疗组(胡黄连苷Ⅱ组),每组10只。胡黄连苷Ⅱ组和阳性对照组动物分别经尾静脉注射胡黄连苷Ⅱ(10mg/kg)和丹参素钠(10mg/kg)干预治疗,阴性对照组和假手术组同步给予等量的0.1mol/L PBS。免疫组化染色检测TLR4和NFκB的表达,TUNEL染色检测细胞凋亡。结果假手术组大鼠皮质、纹状体和海马区脑组织TLR4和NFκB呈微弱表达,TUNEL阳性细胞数量较少,散在分布。阴性对照组大鼠各区脑组织TLR4和NFκB表达(吸光度值,)增强,显著高于假手术组,TUNEL阳性细胞数量较假手术组显著增多(P<0.05)。阳性对照组和胡黄连苷Ⅱ组各区脑组织TLR4和NFκB表达(吸光度值)及TUNEL阳性细胞数量均明显低于阴性对照组(P<0.05),阳性对照组与胡黄连苷组比较差异无统计学意义(P>0.05)。结论胡黄连苷Ⅱ可下调TLR4和NFκB表达而抑制大鼠脑缺血再灌注损伤炎症反应诱导的细胞凋亡。

      Objective To explore the effects of picrodide Ⅱ on the expressions of Toll-like receptor 4 (TLR4) and nuclear transcription factor κB (NFκB) in brain tissue of rat after cerebral ischemic reperfusion (I/R) injury. Methods Ten rats from 60 adult healthy female Wister rats received sham-operation were set as the sham-operative group. Established as middle crerebral I/R model (MCAO/R) by thread tying method,the 30 successfully modeled rats were equally randomized into the negative control group,the positive control group and the treatment group. Besides,rats in the treatment group and the positive control group were respectively intervened with picrodideⅡ (10 mg/kg) and salvianic acid A sodium (10 mg/kg) via caudal vein injection before I/R injury,while rats in the sham-operative group and the negative group were injected with equal volume of 0.1 mol/L PBS. Immunohistochemistry stain was used to determine the expressions of TLR4 and NFκB,and the apoptotic cells were counted by TUNEL-immunofluorescence assay. Results In the sham-operative group,the TLR4 and NFκB expressed weakly with few TUNEL positive cells scattering in the cortex,striatum and hippocampus. As compared with the sham-operative group,TLR4 and NFκB in the negative control group were significantly higher both in absorption ) value and cell number (P<0.05). In the treatment group and the positive control group,the expressions of TLR4 and NFκB and the number of TUNEL positive cells were significantly lower than those in the negative control group (P<0.05),but no significant difference was shown between the two treated groups(P>0.05). Conclusions PicrosideⅡ could down-regulate the expressions of TLR4 and NFκB,and inhibit the inflammatory response induced apoptosis in cerebral I/R injured rats.
