      目的研究维医沙疗对兔膝骨关节炎(osteoarthritis,OA)关节软骨中半胱氨酸蛋白酶(cysteine as-partate-specific proteinase-3,Caspse-3)、B细胞白血病-淋巴瘤-2(B-cell leukemia-lymphoma-2,Bcl-2)、B细胞白血病淋巴瘤-2相关蛋白基因(Bcl-2-associated X protein gene,Bax)及凋亡蛋白表达的影响,为维医沙疗治疗OA提供理论依据。方法新西兰兔22只,右后肢石膏固定的方法建立OA动物模型,其中2只原始模型标本作为对照。其余20只随机分成自由活动组、沙疗组各10只,并将自由活动组健侧作为正常对照组(正常组),沙疗组埋沙治疗20天。免疫组化SP法检测Caspase-3、bcl-2、bax在兔膝关节软骨细胞中的表达。原位末端标记(terminal deoxynucleotidy transferase mediated dUTP nick end labeling,TUNEL)法检测OA软骨细胞凋亡情况。结果活动组及沙疗组阳性细胞个数及光密度(optical density,OD)的表达,Caspase-3、Bax、TUNEL蛋白表达均高于正常组(P<0.01),且沙疗组明显低于活动组(P<0.01);活动组及沙疗组Bcl-2的表达之阳性细胞个数及OD值均低于正常组(P<0.01),沙疗组阳性细胞个数明显高于活动组(P<0.01)。结论维医沙疗能抑制兔OA凋亡细胞Caspase-3、Bax/Bcl-2、TUNEL的表达,可能是维医沙疗的部分抗炎作用机制。
英文摘要:
      Objective To study the effects of sand treatment on cysteine aspartate-specific proteinase-3(Caspse-3),B-cell leukemia-lymphoma-2(Bcl-2),Bcl-2-associated X protein gene(Bax) in the articular cartilage of rabbit knee osteoarthritis(OA),and the apoptotic protein expression,thus providing theoretical evidence for sand treatment in Uyghur medicine.Methods The OA animal model was established by fixing right hind limbs with gypsum in 22 New Zealand rabbits.Of them,2 original model rabbits were taken as the control group,while the rest 20 were randomly divided into a free running group and a sand treatment group,10 in each group.The healthy side of the free running group was taken as the normal control group.The sand treatment was given to those in the sand treatment group for 20 days.The expressions of Caspase-3,Bcl-2,and Bax in the cartilage cells of rabbit knee joint were measured by immunohistochemical assay.The cell apoptosis of chondrocytes was detected using terminal deoxynucleotidy transferase mediated dUTP nick end labeling(TUNEL).Results Compared with the normal control group,the positive cell number,the optical density(OD),the protein expressions of Csapase-3,Bax,and TUNEL were higher in the free running group and the sand treatment group(P<0.01).Besides,they were obviously higher in the free running group than in the sand treatment group(P<0.01).The positive cell number of Bcl-2 and OD value in the free running group and the sand treatment group were both lower than those of the normal control group(P<0.01).The positive cell number was obviously higher in the sand treatment group than in the free running group(P<0.01).ConclusionSand treatment could inhibit the expressions of Caspse-3,Bax/Bcl-2,and TUNEL in the articular cartilage of rabbits’ knee OA,which might be partial mechanisms for anti-inflammation of sand treatment.
