| 用常绿油麻藤种子提取左旋多巴(简称 L-多巴)临床对照观察103例。骨折不愈合及延迟愈合用药组22例,3～6个月达骨性愈合者20例,占90.9%;对照组10例中仅4例6个月内达骨性愈合,占40%。四肢新鲜骨折71例,用药组44例平均临床愈合时间为46.6±12.7天,对照组27例为117.9±66.3天(t=5.96,P<0.001)。动物实验结果也证明L-多巴有加速骨折愈合的作用。 |

| L-dopa,extracted from the seeds of Mucuna Sempervirens Hemsl,is the precursor of transmitter DA.It was applied orally for treatment of non-union,delayed union and fresh fracture in 103 cases of fracture of upper and lower extremities.Of the 32 cases of non-union due to old fracture,22 cases of them were treated by L-dopa,the remaining 10 cases serving as control.The initial dosage of L-dopa was 250mg,three times a day.Three days later,it increased to 500mg.Gastrointestinal symptoms are the main side-effects,yet stimulation to gastrointestinal mucous membrane can be reduced by taking it after meal.Osseous union was found within 3-6 weeks in 20 cases(90.9%)of L- dopa group and in 4 cases(40%)of the control group.For cases of fresh fracture in L-dopa group,the duration of clinical healing was 28-96 days,with an average of 46.6±0.2 days,while that in the control group was 60-231 days, with mean healing duration of 117.9±66.3 days(t=5.96,P<0.001).The osseous tissue healed in the L-dopa group was better and lamellar callus appeared earlier than in the control group. Experimental observation was made in 126 rabbits.Of them 38 radii in 24 rabbits had 1cm defect and 214 radii had 0.3cm defect made by operation.75.38% of the defect radii in L-dopa group and 58.5% in the control group revealed good callus 4 weeks after fracture.Scanning with electron microscope demonstrated that large amount of collagenous fibers appeared in the L-dopa group three weeks after fracture,while the control group showed only fibrocytes.As we know,collagen and bone matrix are the main components of the callus. Our clinical data and experimental observation confirmed Waisman’s view that L-dopa possesses the action of promoting fracture repair and it is a promising drug for treating injuries of bone and joint. |
