电针大鼠双侧"手三里"和"环跳"穴25分钟,随着大鼠痛阈的提高,延桥脑和中脑内5-HT 含量显著升高,5-HIAA 含量无明显变化。但预先腹腔注射 GABA 受体阻断剂荷包牡丹碱不仅能部分阻断电针镇痛效应,也能阻断电针升高大鼠延桥脑和中脑5-HT 的效应,还可以使电针明显升高纹状体5-HT 的水平。降低下丘脑和皮层内5-HIAA的含量。结果说明,在电针镇痛过程中,GABA 能系统起重要作用,而这种作用可能部分通过中枢5-HT 能系统来实现。 |

In the present study,the effect of intraperitoneal injection of bicuculline,a γ-aminobutydc acid(GABA) receptor inhibitor,on analgesic effect of electroacupuncture and the changes in concentrations of 5-hydroxytryptamine(5-HT)and its main metabolite,5-hydroxyindoleacetic acid(5-HIAA),was observed,so as to further ascertain the role of the central GABA system and its regulation on 5-HT metabolism during electroacupuncture analgesia(EAA).After electroacupuncture for 25 minutes at the bilateral Shousanli and Huantiao acupoints of rats,5-HT contents in medulla oblongata and ports as well as midbrain markedly increased,but 5-HIAA contents did not change in the above-mentioned brain regions with elevation of pain threshold.However,a preliminary intraperitoneal injection of bicuculline,not only partially blocked the effect of EAA,but also abolished the effect of 5-HT increase in the medulla-pons and the midbrain,and produced an increase of 5-HT level in striatum and a decrease of 5-HIAA contents in subthalamus and cortex after EA.These results suggest that the GABA system may play an important role in EAA,which may be partially mediated through the central serotonergic system. |
