      目的 :探讨中药当归的主要有效成分当归多糖 (APS)在白血病分化疗法中的应用价值。方法 :应用细胞计数、流式细胞术、形态学、细胞化学、细胞分化免疫表型等现代实验血液学检测技术 ,研究APS对人红白血病细胞株K5 6 2细胞的增殖与分化的作用。结果 :APS对K5 6 2细胞有明显的增殖抑制作用 (P <0 0 5 ) ;经APS诱导后K5 6 2细胞向红系、粒单系细胞方向分化增加 ,联苯胺染色、糖原染色和过氧化物酶染色阳性率、细胞表面分化抗原CD15表达明显增强 (P <0 0 5 ) ;APS诱导后K5 6 2细胞表达C MYC明显降低 (P <0 0 5 )。结论 :APS体外可抑制K5 6 2细胞增殖 ,并诱导其向红系、粒系细胞方向分化 ,是较有开发和应用前景的天然诱导剂。

      To investigate the application value of Angel ic a polysaccharide (APS) on proliferation and differentiation of human erythroleuk emia K562 cells. Methods: The effect of APS in inhibitory proliferation and ind ucing differentiation of human erythroleukemia K562 cells was studied by modern experimental hematologic techniques such as cell counting and culture, flowcytom etry, morphology, cytochemistry and cell differential immune phenotyping. Results: APS could significantly inhibit the proliferation of K562 cells in vitro and prevent the cell from entering the active proliferative phase (P<0 05). After being induced by APS, the differentiatio n of K562 cells to erythrocyte series and granulo monocyte series increased, po sitive rate of benzidine, glycogen and peroxidase stain elevated, and cell surf ace differential antigen CD 15 expression promoted significantly (P <0 05), while C MYC expression of K562 cells induced by APS induction lowered significantly (P<0 05). Conclusion: APS could not only inhibit the proliferation of K5 62 cells in vitro, but also induce the differentiation of K562 cells toward eryt hrocyte and granulocyte series. It may be a natural inducer with promising prosp ect of development and application.
