      目的 :研究不同浓度的加味玉屏风汤对地塞米松磷酸钠 (Dex p)诱导的小鼠胸腺细胞凋亡和胸腺细胞自发凋亡的影响。方法 :通过体外细胞培养 ,采用琼脂糖凝胶电泳、荧光显微镜检测手段 ,观察加味玉屏风汤作用前后对胸腺细胞凋亡程度的影响。结果 :胸腺细胞既可发生自发凋亡 ,又可由Dex p诱导凋亡 ,细胞凋亡率与Dex p呈浓度依赖关系 ,最佳诱导浓度为 10 - 7mol/L。加味玉屏风汤 (2 0 0~ 4 0 0 μg/ml)可不同程度地抑制由Dex p(10 - 7mol/L)诱导的胸腺细胞凋亡 (P <0 0 1) ;而低浓度 (<10 0 μg/ml)的加味玉屏风汤则对Dex p(10 - 7mol/L)诱导的胸腺细胞凋亡无明显影响 (P >0 0 5 ) ;加味玉屏风汤 2 0 0、4 0 0 μg/ml可使胸腺细胞的自发凋亡率明显降低 (P <0 0 1)。结论 :不同浓度加味玉屏风汤 (2 0 0~ 4 0 0 μg/ml)可不同程度抑制胸腺细胞的自发凋亡及由Dex p(10 - 7mol/L)诱导的凋亡。
英文摘要:
      Objective: To explore the effect of Jiawei Yupingfeng Decoction (JYD) in different concentration on thymocyte apoptosis either spontaneously occurred or induced by Dexamethasone sodium phosphate (Dex p). Methods: Conditions of thymocyte (cultured in vitro) apoptosis before and after treatment with JYD were observed by agarose gel electrophoresis and fluorescence microscopy. Results: Thymocyte apoptosis could occur spontaneously or induced by Dex p in concentration dependent manner, with the optimal concentration of 10 -7 mol/L. JYD in concentration of 200~400μg/ml could inhibit the Dex p (10 -7 mol/L) induced thymocyte apoptosis to different degree (P<0 01), while JYD in low concentration (<100μg/ml) showed no effect on it. JYD of 200~400μg/ml could lower the spontaneous apoptosis of thymocyte significantly (P<0 01). Conclusion: JYD in different concentration (200~400μg/ml) could inhibit the thymocyte apoptosis, either occurred spontaneously or induced by Dex p, to different degree.
