目的 :探讨牛珀至宝微丸对大鼠局灶性脑缺血后脑组织一氧化氮合酶 (NOS)的作用。方法 :采用大脑中动脉线栓法造成局灶性脑缺血模型 ,应用免疫组织化学技术检测脑组织NOS 3种亚型 (eNOS、nNOS、iNOS)的表达 ,观察牛珀至宝微丸对脑缺血后NOS 3种亚型表达的影响 ,并进行神经病学评分。结果 :牛珀至宝微丸组的缺血侧大脑皮层nNOS、iNOS表达显著低于缺血对照组 (P <0 0 5 ,P <0 0 1) ,eNOS表达高于缺血对照组 (P <0 0 5 ) ,并可减轻神经损伤症状 (P <0 0 5 )。结论 :牛珀至宝微丸能下调脑缺血所致nNOS、iNOS的异常表达 ,上调eNOS表达 ,这可能是其治疗缺血性脑血管病的机理之一。 |

Objective: To study the effect of Niupo Zhibao pellet (NZP) on nitric oxide synthase (NOS) in brain of focal cerebral ischemic rats. Methods: Focal ischemic model of rats established by inserting thread in cerebral medium artery was used in this experiment. Effect of NZP on expression of 3 subtypes of NOS (eNOS, nNOS and iNOS) in brain of rats was determined by immunohistochemical technique and neurological evaluation was carried out. Results: As compared with the ischemic control group, levels of nNOS and iNOS in the NZP group were lower (P<0 05 and P<0 01) and eNOS higher (P<0 05). And the symptoms of nerve damage were reduced in the NZP group (P<0 05). Conclusion: NZP could down regulate the abnormal expression of nNOS and iNOS induced by cerebral ischemia, and up regulate that of eNOS, this may be one of the mechanisms of its therapeutic effect. |
