      目的 :观察补肾中药对垂体促性腺激素释放激素 (GnRH)受体及其基因表达的影响 ,探讨补肾中药调整下丘脑 -垂体 -性腺轴功能的作用环节及作用机理。方法 :将青春期大鼠分为对照组、滋阴泻火组和益肾填精组 ,采用免疫组织化学方法检测GnRH受体的蛋白表达水平 ,采用原位杂交组织化学方法检测GnRH受体mRNA表达水平。结果 :给予滋阴泻火中药合剂后 ,大鼠垂体GnRH受体mRNA表达水平显著下调 ,GnRH受体蛋白合成减少 ;而给予益肾填精中药合剂后 ,大鼠垂体GnRH受体mRNA表达水平显著上调 ,GnRH受体蛋白合成增多。结论 :不同补肾中药可通过改变垂体GnRH受体及其mRNA表达水平 ,调节下丘脑 -垂体 -性腺轴的功能。

      Objective: To observe the effect of Chinese drug for reinforcing Shen (CRS) on the expression of gonadotropin releasing hormone (GnRH) receptor and its mRNA, in order to explore the mechanism of regulatory effect of CRS on function of hypothalamic pituitary gonadal axis. Methods: Female SD rats were randomly distributed into three groups, the control group, the tested group A (treated with drugs for nourishing Yin and purging Fire) and tested group B (treated with drugs for supplementing Shen essence). Integrated optic density of GnRH receptor and its mRNA in rat′s hypophysis were determined by immunohistochemistry, in situ hybridization method and computer image processing respectively. Results: After treatment, in the tested group A, GnRH receptor mRNA expression was down regulated significantly and synthesis of GnRH receptor protein reduced, while an opposite (up regulation) result was found in the tested group B. Conclusion: Different CRS could modulate the function of the hypothalamic pituitary gonadal axis through regulating the expression of GnRH receptor and its mRNA in hypophysis.
