      目的探讨柴胡提取物调节心肌细胞凋亡的作用机制。方法以柯萨奇病毒嗜心肌株(CVB3m)腹腔注射诱导BALB/c小鼠病毒性心肌炎模型,将实验小鼠分为正常对照组、模型组、柴胡提取物组,柴胡提取物组每天予0·2ml/10g的浓度为2mg/ml的柴胡提取物灌胃,正常对照组、模型组予以等量生理盐水灌胃,于造模后第10天处死,采用TUNEL法检测心肌细胞凋亡,采用免疫组化、RT-PCR法检测各组小鼠心肌中Fas、Bcl-2、Caspase3表达。结果正常组小鼠心肌未检测到凋亡的心肌细胞,Fas、Bcl-2、Caspase3均有不同程度表达,模型组可见大量心肌细胞凋亡,且Fas、Bcl-2、Caspase3表达均强于对照组(P<0·01),柴胡提取物组心肌细胞凋亡明显减少,与模型组比较差异显著(P<0·01),Fas、Caspase3表达明显降低,Bcl-2表达轻度升高。结论柴胡提取物可以抑制病毒性心肌炎时心肌细胞凋亡,明显下调Fas及Caspase3表达,轻微上调Bcl-2表达。

      ObjectiveTo explore the mechanism of Radix Bupleuri (RB) extract in regulating cardiomyocyte apoptosis. MethodsThe BALB/c mouse inoculated with the cardiomyocytophilic strain Coxsackie virus B3m (CVB3m) by peritoneal injection was adopted as viral myocarditis model. The experimental mice were divided into the normal control (A), the model (B) and the RB (C) groups. Mice in Group C were given 0.2 ml/10g of RB extract, 2 mg/ml in concentration by gastrogavage once daily, while equal volume of normal saline was given by gastrogavage to mice in Group A and B. All the mice were sacrificed on the 10 th day after modeling. The cardiomyocyte apoptosis was detected by TDT/Dutp nick end labelling (TUNEL). The expression of Fas Bcl-2, and Caspase3 protein in myocardium was determined by immunohistochemistry and reverse-transcription polymerse chain reaction (RT-PCR). ResultsThe apoptotic cell was not found in Group A, but Fas, Bcl-2 and Caspase3 were expressed in different degree. In Group B, a lot of apoptotic cells could be seen, with expression of Fas, Bcl-2 and Caspase3 stronger than those in Group A (P<0.01). Compared with Group B, the apoptotic cardiomyocyte in Group C was significantly reduced (P<0.01), with the expression of Fas and Caspase3 significantly lowered but expression of Bcl-2 increased slightly. ConclusionRB extract could inhibit the cardiomyocyte apoptosis in animal with viral myocarditis, obviously down-regulate the expression of Fas and Caspase3, and slightly up-regulate Bcl-2 expression.
