目的通过磁共振,动物神经行为学评分以及脑组织超微组织观察判断栀子苷、胆酸及其配伍对局灶缺血性大鼠的治疗作用。方法用线栓法建立大鼠脑缺血再灌注模型,应用磁共振弥散加权成像(MRDWI)技术于造模后缺血再灌注3h综合评价组分配伍的治疗作用,神经功能检查法对脑缺血后动物进行行为测评,病理及超微组织观察作为药效评价辅助手段,准确界定不同组分配伍的功效作用。结果与模型组比较,各治疗组病灶的表现弥散系数值及平均弥散系数值有升高趋势;栀子苷加胆酸组病灶周边区差异有显著性(P<0.05)。结论栀子苷加胆酸组分配伍具有脑神经保护作用,磁共振技术结合行为学评分及脑组织超微组织观察是评价神经保护剂的有效手段。 |

ObjectiveTo estimate the therapeutic effect of single or combined use of jasminoidin and cholalic acid on focal cerebral ischemia rat with magnetic resonance-diffusion-weighted imaging (MR-DWI) technique, ultra-microscopy, and neuro-behavior scoring. MethodsThe model of cerebral ischemia-reperfusion injury was induced by string method. Three hours after reperfusion, MR-DWI was applied with ultra-microscopy and neuro-behavior test to give evaluation on cerebral ischemic rats, and pathologic, ultramicroscopic observation of tissue were taken as adjuvant measures to comprehensively evaluate the pharmacological effect on ischemia-reperfusion rats and delimit the efficacy of the two different components and their combination. ResultsCompared with the model group, ADC and DCavg values of the foci in all the treated groups had the incrensing trend. There was significant difference arund the foci in the group of combined use of jasminoidin and cholalic acid (P<0.05). Conclusion Combined use of jasminoidin and cholalic acid had protective effects on nerve and brain. MR-DWI technique accompanied with ultramicroscopic observation of tissues and neuro-behavior test is an effective method for evaluating the effect of neuro-protective agent. |
