目的动态观察红花注射液对大鼠肺血栓栓塞症(PTE)中血管内皮细胞间黏附分子-1(ICAM-1)和P-选择素的影响。方法将Wistar大鼠随机分为正常组、模型组和治疗组,用颈静脉注入自体血凝块制备动物模型。治疗组在模型建立后立即自鼠尾静脉注射红花注射液2ml/kg,每天1次,共5天。各组大鼠分别于栓塞后1、3、24、72、120h处死,行病理观察;采用免疫组化和原位杂交的方法检测肺血管内皮I-CAM-1和P选择素蛋白及mRNA的表达水平。结果与正常组比较,模型组大鼠肺栓塞后病理可见肺动脉血栓形成,炎性反应明显;肺血管内皮ICAM-1蛋白及mRNA表达在栓塞后3h开始增高(P<0·01),而P-选择素蛋白及mRNA表达在栓塞后1h开始增高(P<0·01)。治疗组大鼠病理损伤减轻,肺血管内皮ICAM-1和P-选择素水平均下降(P<0·01)。结论红花注射液可减轻大鼠PTE时的肺损伤,这种作用可能是通过下调血管内皮上的黏附分子来实现的。 |

ObjectiveTo observe dynamically the express ions of intercellular adh esion molecule-1 (ICAM-1) and P-selectin (Ps) in the vascular endothelium i n rat s with pulmonary thromboembolism (PTE) and the effect of Safflower Injection ( S I). MethodsRats were randomly divided into the normal g roup, the model group a nd the treatment group. PTE model was induced by intravenous injection of auto- b lood clots, and SI was injected intravenously immediately after modelling with 2 ml/(kg·d) for 5 days to the rats in the treatment group. Animals were sacrific ed in batches at the 1st, 3rd, 24th, 72nd and 120th h after embolization to observe the pathological changes and detect the protein and mRNA expressions of ICAM-1 and Ps in pulmonary vascular endothelium by immunohistochemistry and in situ hyb ridization respectively. ResultsPathological observatio n showed obvious emboli sm in pulmonary arteries and inflammatory reaction after modelling, which was ab ated after SI treatment. ICAM-1 and Ps expressions were elevated at the 3rd and th e 1st h after embolization respectively (both P<0.01), which a lso were abated in the treatment group. ConclusionSI may alleviate pulmona ry injury in PTE rats by down-regulating the expressions of ICAM-1 and Ps. |
