      目的用血清药理学的方法研究炙甘草汤含药血清对兔心肌细胞瞬间外向钾电流(Ito)的影响。方法进行单个心室肌细胞的分离和膜片钳全细胞记录,将心室肌细胞分为对照组、空白血清组及5%、10%、20%、40%含药血清组,分别以普通细胞外液及加入上述不同浓度血清的细胞外液进行灌流,检测不同浓度含药血清对Ito的影响。结果含药血清可抑制Ito,5%、10%、20%、40%含药血清分别将Ito峰值(PA/PF)从(16·1±1·4)降至(13·9±1·5)、(11·8±1·9)、(8·3±1·5)、(8·2±1·2),洗脱后其作用可消除。结论炙甘草汤含药血清可抑制Ito,且呈浓度依赖性作用增强,可能是炙甘草汤抗心律失常作用的机理。
英文摘要:
      ObjectiveTo investigate the effect of Zhigancao Decoction (ZD) containing serum on transient outward potassium current (I_ to ) in isolated single ventricular myocyte of rabbits. MethodsSingle rabbit ventricular myocytes were isolated and divided into 6 groups, the normal group, the non-drug-containing serum group, and the ZD groups, they were treated with normal extracellular fluid alone or with ZD containing serum in different concentrations (5%, 10%, 20% and 40%) respectively by perfusion. Whole-cell patch clamp recording technique was used to observe the changes of I_ to . ResultsAfter treated with 5%, 10%, 20% and 40% ZD containing serum, the I_ to was inhibited and its peak value dropped from (16.1±1.4) PA/PF to (13.9± 1.5) PA/PF, (11.8±1.9) PA/PF, (8.3±1.5) PA/PF and (8.2±1.2) PA/PF respectively, and the effect could be eliminated by elution. ConclusionZD containing serum could inhibit the I_ to in isolated ventricular myocytes of rabbits in a concentration-dependent manner, which may be the mechanism of antiarrhythmic action of ZD.
