      目的观察慢性心理应激对血管内皮功能障碍大鼠的影响及通心络的干预作用,并探讨其机制。方法 40只雄性Wistar大鼠随机分为正常对照组、内皮功能障碍组(HCY组)、心理应激组(模型组)、通心络组,每组10只;用3%高蛋氨酸饮食复制内皮功能障碍模型,利用重复束缚法对内皮功能障碍大鼠进行心理刺激,并用通心络(1.2g/kg)灌胃给药。放免法检测血浆内皮素(ET)、血浆血管紧张素Ⅱ(AngⅡ)、血清皮质酮(CORT);硝酸还原酶法检测血清一氧化氮(NO);透射电镜观察主动脉组织内皮细胞超微结构变化;ELISA法检测血清去甲肾上腺素(NE)、肾上腺素(E)水平。结果与HCY组血浆ET[(161.70±13.96)pg/mL],血清NO[(26.82±13.03)μmol/L]比较,模型组大鼠血浆ET[(178.25±21.85)pg/mL]明显升高(P<0.05),血清NO[(24.91±9.95)μmol/L]无明显下降(P>0.05);CORT、NE、E明显升高(P<0.05);内皮细胞超微结构明显损伤。与模型组比较,通心络组血浆ET[(154.74±13.27)pg/mL]、AngⅡ、CORT、NE、E明显降低(P<0.05,P<0.01),血清NO[(34.44±18.35)μmol/L]明显升高(P<0.05);内皮细胞超微结构显著改善。结论慢性心理应激可明显加重血管内皮功能障碍大鼠的内皮损伤,通心络具有保护血管内皮结构和功能的作用。

      Objective To observe the effect of chronic psychological stress on vascular endothelial dysfunction rats and to explore the intervention and mechanism of Tongxinluo (TXL) on it. Methods Forty male Wistar rats were randomly divided into the normal control group (with no modeling),the endothelial dysfunction group (the HCY group),the psychological stress group (the model group),and TXL group,ten in each group. Rats in the latter three groups were fed with 3% high methionine diet to duplicate vascular endothelial dysfunction (VED) model. In addition,chronic psychological stress was applied in VED rats using repeated binding method. TXL at the dose of 1.2 g/kg body weight was given by gastrogavage. The plasma endothelin (ET) and angiotensin Ⅱ (Ang Ⅱ),serum cortisone (CORT) were detected by radioimmunoassay. The serum nitric oxide (NO) was detected by nitrate reductase. Ultrastructural changes of aortic endothelial cells were observed by transmission electron microscope. Serum levels of norepinephrine (NE) and epinephrin (E) were detected by enzyme linked immunosorbent assay (ELISA). Results Compared with the plasma ET level and the serum NO level in the HCY group (161.70±13.96 pg/mL and 26.82±13.03 μmol/L),the plasma ET level obviously increased (178.25±21.85 pg/mL) (P<0.05) and the serum NO level decreased (24.91±9.95 μmol/L,P>0.05),levels of CORT,NE,and E obviously increased in the model group (all P<0.05). Ultrastructural changes of aortic endothelial cells were obviously injured. Compared with the model group,the plasma ET level (154.74±13.27 pg/mL),Ang Ⅱ,CORT,NE,and E obviously decreased (P<0.05,P<0.01),the serum NO level obviously increased (34.44±18.35 μmol/L,P<0.05). Ultrastructural changes of aortic endothelial cells were obviously improved. Conclusions Chronic psychological stress could obviously aggravate endothelial injury in VED rats. TXL showed protection on the vascular endothelial structure and function.
