| 目的观察补肾序贯方结合软坚化瘀方对子宫内膜异位症(endometriosis,EMS)大鼠异位内膜基质金属蛋白酶-2(MMP-2)的调节作用,探讨子宫内膜异位症的发病机制与治疗机制。方法建立EMS大鼠模型,将造模成功的SD雌性大鼠随机分成混合序贯组(补肾序贯结合软坚化瘀组)、软坚化瘀组和模型组,并设正常对照组。喂药4周后,择动情期处死大鼠,镜下观察各组大鼠异位或在位内膜组织中MMP-2的分布及染色强度;采用链霉素抗生物素蛋白-过氧化酶连接(SP)法测定MMP-2蛋白的表达水平。结果模型组大鼠异位内膜组织中MMP-2蛋白表达明显高于正常对照组(P<0.01);与模型组比较,混合序贯组、软坚化瘀组大鼠异位内膜组织中的MMP-2蛋白表达均有所下降(P<0.05);从MMP-2的分布及阳性蛋白颗粒染色上的变化中观察,混合序贯组的治疗效果最明显。结论子宫内膜异位症的发生与MMP-2活性升高有关,补肾序贯和软坚化瘀法合用较单纯使用软坚化瘀法能够更有效地抑制EMS大鼠异位内膜组织中的MMP-2活性,两法合用方能改善内分泌环境,通过降低异位内膜"侵袭"能力达到治疗作用。 |

| Objective To observe the effect of the combination of Bushen Xuguan Recipe(BXR) and Ruanjian Huayu Recipe(RHR) on the matrix metalloproteinases-2(MMP-2) expression of rats with endometriosis(EMS),and to study the pathogenesis and mechanism of EMS.Methods The EMS rat model was established.Successfully modeled female SD rats were randomly divided into the combination group(BXR+ RHR),the RHR group,and the model group.And a normal control group was set up.After 4 weeks of medicinal treatment,rats were sacrificed in the oestrus cycle.MMP-2 distribution and the staining density in ectopic and eutopic endometrium of the rats were observed under microscope.The MMP-2 expression was determined by immunohistochemical assay(SP method).Results The MMP-2 expression in the ectopic endometrial tissues of rats in the model group were significantly higher than that of the normal control group(P<0.01).Compared with the model group,the MMP-2 expression decreased in the combination group and the RHR group(P<0.05).The therapeutic effect of the combination group was most obvious by observing the distribution of MMP-2 and changes of positive staining.Conclusions The occurrence of EMS was correlated with increased activities of MMP-2.The combination method could inhibit the activities of MMP-2 in the ectopic endometrial tissues of EMS rats more effectively than RHR alone,which might be achieved through improving the endocrine environment,and reducing the "invasion" capability of the ectopic endometrium. |
