      目的观察补气通络解毒方对胰腺癌小鼠模型HIC1基因甲基化的影响。方法将30只裸鼠随机分为正常组、模型组及治疗组,每组10只,采用癌细胞皮下种植法造模,治疗组给予补气通络解毒方(60 g生药/kg,3 mL/100 g)灌胃,正常组和模型组给予等体积生理盐水灌胃,连续干预14天后,处死裸鼠,取瘤组织,以巢式甲基化特异性聚合酶链反应(nested methylation-specific polymerase chain reaction,N-MSP)法检测各组标本的HIC1基因甲基化程度。结果模型组HIC1基因甲基化比率明显高于正常组(P<001),而治疗组与模型组比较,HIC1基因甲基化程度降低(P<001)。结论补气通络解毒方能降低模型小鼠HIC1基因甲基化程度,从而发挥治疗胰腺癌的分子生物学效应。
英文摘要:
      ObjectiveTo observe the effect of Buqi Tongluo Jiedu Recipe (BTJR) on HIC1 methylation of pancreatic cancer mouse model. MethodsTotally 30 nude mice were randomly divided into the normal group, the model group, and the treatment group, 10 in each group. The model was induced by cancer cell subcutaneous planting method. Mice in the treatment group were administered with BTJR (60 g crude drugs/kg, 3 mL/100 g) by gastrogavage. Equal volume of normal saline was given to those in the normal group and the model group by gastrogavage. All the intervention lasted for 14 successive days. The mice were sacrificed to death. Their tumor tissues were taken out. The HIC1 gene methylation level of each specimen was detected by nested methylation-specific polymerase chain reaction (N-MSP) method. ResultsCompared with the model group, the HIC1 gene methylation level was lower in the treatment group (P<0.01). Compared with the model, the HIC1 gene methylation level was even lower in the mormal group (P<0.01). ConclusionBTJR could reduce HIC1 gene methylation degree of model mice, thus playing a role of molecular biologic effect in treating pancreatic cancer.
