      目的 运用非靶向代谢组学技术分析冠心病痰湿证、血瘀证和痰瘀互结证3种证候的潜在生物标志物,从分子生物学角度为冠心病的中医辨证分型提供依据。方法 收集120例冠心病稳定期患者(痰湿证38例,血瘀证18例,痰瘀互结证64例)及51名健康人的血液样品,采用超高效液相色谱-四极杆-静电场轨道阱质谱(UHPLC-Q-Orbitrap HRMS)技术对样品进行检测,利用Progenesis QI和Simca-P软件对数据进行多元统计分析及内源性生物标志物的筛查,通过MetaboAnalyst数据库分析代谢通路。结果 共得到10个潜在的生物标志物,其中N-乙酰鸟氨酸和月桂酸是冠心病血瘀证潜在的生物标志物,1,1-二氯乙烯环氧化物是冠心病痰瘀互结证潜在的生物标志物,其余6个为冠心病共性差异代谢物,涉及视黄醇代谢、精氨酸和脯氨酸代谢等9条代谢通路。结论 冠心病痰湿证、血瘀证和痰瘀互结证3种证候之间有共性代谢物及差异性代谢物,对于冠心病中医证候辨证具有重要的参考价值。

      Objective In this study,non-targeted metabolomics was used to analyze the potential biomarkers of 3 syndromes of coronary heart disease (CHD): phlegm-dampness syndrome, blood stasis syndrome, phlegm and blood stasis syndrome,and provide theoretical basis for Chinese medicine syndrome differentiation of CHD from the perspective of molecular biology. Methods The blood samples of 120 patients with stable CHD (38 cases of phlegm-dampness syndrome,18 cases of blood stasis syndrome,64 cases of phlegm and blood stasis syndrome) and 51 healthy participants were collected. Ultra-high performance liquid chromatography-quadrupole-orbitrap high resolution mass spectrometry technology was used to detect each group of samples,Progenesis QI and Simca-P software were used for multivariate statistical analysis,and metabolic pathways were analyzed through the MetaboAnalyst database. Results A total of 10 potential biomarkers were identified,among which N-Acetylornithine and Dodecanoic acid were potential markers of blood stasis syndrome,1,1-Dichloroethylene epoxide was potential biomarkers of phlegm and blood stasis syndrome,and the other 6 were common differential metabolites of CHD,involving 9 metabolic pathways such as retinol metabolism,arginine and proline metabolism. Conclusion There are common and different metabolites among the 3 syndromes of phlegm-dampness syndrome, blood stasis syndrome,phlegm and blood stasis syndrome, which are of important reference value for the study of CHD Chinese medicine syndrome .
