      目的 研究大蒜素调控核因子κB受体活化因子(RANK)对破骨细胞分化成熟的影响。方法 诱导小鼠骨髓间充质干细胞(BMSCs)分化破骨细胞,分为对照组及实验组,两组采用30 ng/mL巨噬细胞集落刺激因子(M-CSF)及60 ng/mL RANKL培养,实验组第1、3、7天进行大蒜素(20 μg/mL)干预。采用CCK-8检测两组细胞的增殖情况,通过抗酒石酸酸性磷酸酶(TRAP)染色及定量检测TRAP酶活性,通过RT-PCR检测RANK mRNA表达量。结果 与本组第1天比较,两组第3天破骨细胞增殖均降低(P<0.01),TRAP酶活性增加(P<0.01),实验组第3天RANK mRNA表达降低(P<0.05);与本组第3天比较,第7天两组破骨细胞增殖能力增高(P<0.01)。与对照组比较,实验组第1天RANK mRNA表达更高(P<0.05),第3、7天破骨细胞增殖能力及RANK mRNA表达降低(P<0.05,P<0.01),第7天TRAP酶活性降低(P<0.01)。结论 大蒜素对破骨细胞分化成熟具有显著抑制作用,其对破骨细胞胞的增殖、RANK mRNA表达、破骨细胞活性作用均有抑制作用,且呈时间依赖性。

      Objective To study the effect of allicin regulating receptor activator of nuclear factor-κB(RANK) on osteoclast differentiation and maturation.Methods Mouse bone marrow mesenchymal stem cells (BMSCs)were induced to make osteoclast differentiation,then the cells were divided into control group and experimental group.At 1,3,7 days,the two groups were cultured by 30 ng/mL macrophage-stimulating factor(M-CSF)and 60 ng/mL RANKL,the experimental group was received allicin (20μg/mL) intervention.The proliferation of cells in the two groups was detected by CCK-8,the TRAP enzyme activity was detected by tartrate-resistant acid phosphatase(TRAP) staining and quantitative detection,and RANK mRNA expression was detected by RT-PCR.Results Compared with the 1st day,the proliferation of osteoclasts in the two groups decreased (P<0.01), the activity of TRAP enzyme increased(P<0.01),and the expression of RANK mRNA in the experimental group decreased on day 3(P<0.05).Compared with the 3rd day, the proliferation ability of osteoclasts in the two groups increased on the 7th day(P<0.01).Compared with the control group,RANK mRNA expression in the experimental group was higher on 1st day(P<0.05),osteoclast proliferation and RANK mRNA expression decreased on day 3 and 7 (P<0.05),P<0.01),and TRAP enzyme activity decreased on 7th day(P<0.01).Conclusion Allicin has significant inhibitory effect on osteoclast differentiation and maturation,and has inhibitory effects on osteoclast cell proliferation,RANK mRNA expression and osteoclast activity in a time-dependent manner.
