      目的 :观察雷公藤多甙对急性移植物抗宿主病时小鼠脾、皮肤、肺、肠组织中T细胞及其亚群 (CD3+ 、CD4 + 、CD8+ 细胞 )的影响 ,探讨雷公藤多甙对急性移植物抗宿主病的作用及机理。方法 :用直线加速器排空受鼠骨髓后注入供鼠骨髓与脾淋巴细胞的混合液 ,复制出急性移植物抗宿主病 (aGVHD)模型。观察雷公藤多甙、雷公藤多甙加环孢素A(CsA)、CsA加甲氨蝶呤对各组小鼠脾CD3+ 、CD4 + 、CD8+ 细胞 (流式细胞仪检测 ) ,皮肤、肺、肠组织CD3+ 、CD4 + 、CD8+ 细胞浸润情况 (应用免疫组化技术检测 )的影响。结果 :雷公藤多甙组小鼠脾、皮肤、肺组织CD3+ 、CD4 + 、CD8+ 细胞明显低于异基因移植组 (P <0 0 5 ) ,而小肠组织CD3+ 、CD4 + 、CD8+ 细胞变化不明显 (P >0 0 5 )。结论 :雷公藤多甙可降低脾T细胞及亚群数量 ,减少皮肤、肺的T细胞浸润 ,具有明显的抗aGVHD作用。

      Objective: To observe the effect of Glucosidorum tripterygii tororum (GTT) on T lymphocyte and its subsets in mice with acute graft versus host disease (aGVHD). Methods: The recipient mice, after their bone marrow had been evacuated by radiation with linear accelerator, were given the mixture of hone marrow and splenic T lymphocyte obtained from donor mice to duplicate aGVHD model. The effect of GTT, GTT+Cyclosporin A and CsA+Methotrexate on T lymphocyte subsets (CD 3 +, CD 4 + and CD 8 +) in spleen was determined by flow cytometer, and those on lymphocyte infiltration in skin, lung and intestine was detected with immunohistological technique. Results: CD 3 +, CD 4 + and CD 8 + cells in spleen, skin and lung in mice of the GTT treated groups were significantly lower than those in the model group (P<0 05), while in intestinal tissue, the difference was insignificant (P>0 05). Conclusion: GTT could reduce the T lymphocyte and its subsets in spleen and the T lymphocyte infiltration in skin and lung, showing evident anti aGVHD effect.
