      目的 :研究生脉散皂苷部位对培养的大鼠单个心肌细胞内Ca2 +浓度的影响。方法 :以Fluo 3AM负载心肌细胞 ,采用激光扫描共聚焦显微镜检测单个细胞内Ca2 +浓度的变化。结果 :生脉散皂苷部位10 μg/ml可明显增加心肌细胞内Ca2 +浓度 ,并使其维持在高水平 (P <0 0 1) ,随后加入高Ca2 +和高K+对细胞内Ca2 +浓度无明显影响。生脉散皂苷部位 10 0 μg/ml诱导心肌细胞内Ca2 +浓度迅速明显增加后又迅速下降至静息水平 (P <0 0 1) ,外加高Ca2 +和高K+不能刺激细胞内Ca2 +浓度明显增加。结论 :生脉散皂苷部位对心肌细胞内Ca2 +浓度变化的影响与其浓度有关。
英文摘要:
      Objective: To study the effects of saponins extracted from Shengmai Powder (SSMP) on Ca 2+ level in cultured single neonatal rat myocardial cells. Methods: Myocardial cells were cultured and loaded with Fluo 3 AM. The change of Ca 2+ level in each cell was determined with laser scanning confocal microscopy. Results: SSMP 10μg/ml caused an obvious elevation of Ca 2+ level (P<0 01), which maintained on a level that was higher than the resting level. No obvious change was observed on Ca 2+ level, followed by the addition of high concentrated Ca 2+ and K +. SSMP 100μg/ml induced a rapid and obvious elevation of Ca 2+ level (P<0 01), which rapidly dropped down to the resting level. Extracellular high level of Ca 2+ and K + had no influence on intracellular Ca 2+ level. Conclusion: The effect of SSMP on Ca 2+ in cultured myocardial cells is related to the concentration used.
