      目的在爪蟾卵母细胞中建立人低密度脂蛋白受体(lowdensitylipoproteinreceptor,LDL- R)表达系统,并研究姜黄素降脂作用的分子机理。方法将含人LDL R的表达质粒p3 7LDL导入细胞核中,用免疫荧光、配体荧光以及免疫胶体金标记透射电镜等方法检测细胞膜上LDL- R的表达情况;用不同浓度的姜黄素对爪蟾卵母细胞人LDL- R基因的表达进行干预,并用酶联免疫吸附分析法测定LDL- R的表达量。结果人LDL -R基因在爪蟾卵母细胞膜上得到表达;姜黄素具有增强其表达的作用,且有明显的量效关系。结论姜黄素降血脂和抗动脉粥样硬化的作用途径之一可以是通过促进LDL- R基因的表达,增加细胞对低密度脂蛋白胆固醇的吸收,从而降低血清胆固醇。
英文摘要:
      ObjectiveTo investigate the molecular mechanism of curcumin in reducing blood lipids by establishing gene expression system of human low density lipoprotein receptors (LDL-R) in Xenopus Laevis oocytes (XLO). MethodsThe expression of LDL-R on cytomembrane was determined using immuno-fluorescent, ligand-fluorescent and immune colloidal gold techniques after human LDL-R containing p3.7 LDL plasmid was led into nucleus. And the expression of LDL-R gene in XLO was quantitatively determined by ELISA after being interfered with different concentrations of curcumin. ResultsThe human LDL-R gene could be expressed on XLO, which could be significantly enhanced by curcumin in a dose-dependent manner. ConclusionOne of the paths of curcumin in reducing blood lipids and anti-atherosclerosis was improving LDL-R gene expression and increasing the LDL-cholesterol absorption of cells.
